My laboratory is focused on understanding two facets of lung endothelial cell biology: 1) structural and functional adaptations in lung to heart failure and 2) factors which dictate the spatial heterogeneity in endothelial targets for acute lung injury. Currently funded work relates to identifying a role for the vanilloid transient receptor potential channel TRPV4 in regulation of calcium entry into endothelial cells, which impacts endothelial permeability. Further, we are evaluating the hypothesis that endothelial injury “fingerprints” in the lung are dictated by the spatial distribution of specific TRP channels. This work utilizes a variety of techniques ranging from specific measures of endothelial permeability in whole lung, microscopy and vascular corrosion casting, immunohistochemistry, and knockout or transient knockdown of TRPV4 in lung. |
